I was playing Buzz Video Game and I went through everything and I was defeating the enemies in the room before getting to beat Zurg and before the floor started to lower, I decided to save the game and when I started the game today, it began at the start where Buzz was flying through the air. Do you have to play the whole level in one day? Then why do they let you save and exit when it doesn't let you save your progress in a level?

Also, how do you open up new levels (I'm not talking about the Toy Box) in the main story mode?

You have to play threw each level whole in one run through. They let you save the game in order to save the capsules, special cards, and aliens that you collect that are hidden throughout each level. Also, in order to open up different level you would have to beat each level as it opens up starting with the Buzz Game.
